PanNDA: New Protein Atlas Decodes Complexity of Neurodegenerative Diseases

Now, a new resource called the pan-neurodegeneration atlas, or PanNDA, is offering scientists a more detailed map of that web, potentially accelerating the search for effective treatments. Developed by a collaborative team led by researchers at St. Jude Children’s Research Hospital, PanNDA is a comprehensive survey of what are known as “proteomes” from various neurodegenerative diseases. This means it catalogs the levels of proteins, how they are modified, and how they interact within cells affected by these conditions.

Decoding the Molecular Signatures of Neurodegeneration

The study, published today in the journal Cell, represents a significant step forward in understanding the shared biological mechanisms underlying these devastating illnesses. For years, researchers have recognized that neurodegenerative diseases, despite their distinct clinical presentations, often share overlapping molecular signatures. PanNDA aims to decode these similarities and differences, providing a protein-based perspective that complements existing genetic and other types of data. A proteome is the entire set of proteins expressed by a cell or organism at a particular time. Analyzing proteomes allows scientists to move beyond the genetic blueprint (the genome) and understand what is actually happening within cells. Proteins can be modified in numerous ways – phosphorylation, ubiquitination, and glycosylation are just a few examples – and these modifications can dramatically alter their function. PanNDA doesn’t just identify which proteins are present, but likewise how they are modified and how they interact with each other. Building the Atlas: Methods and Scope

The PanNDA project involved analyzing tissue samples from multiple brain regions of individuals with Alzheimer’s disease, Parkinson’s disease, frontotemporal dementia, and other neurodegenerative conditions. Researchers used mass spectrometry, a powerful technique that allows them to identify and quantify proteins, to create a detailed snapshot of the proteome in each sample. The development of PanNDA has several potential implications. First, it could lead to the identification of biomarkers – measurable indicators of disease – that could be used to diagnose neurodegenerative diseases earlier and more accurately. Currently, diagnosis often relies on clinical symptoms, which can be subtle and nonspecific in the early stages of the disease. Second, the atlas could aid researchers identify new therapeutic targets. By pinpointing the key proteins and pathways that are disrupted in neurodegenerative diseases, scientists can develop drugs that specifically target these abnormalities.

Researchers are already using PanNDA to explore potential drug candidates and to understand why some individuals are more resistant to neurodegenerative diseases than others. The Human Protein Atlas, a related resource, provides a wealth of information about protein expression in normal tissues, which can be used to compare and contrast with the findings from PanNDA. This comparative approach can help identify proteins that are specifically altered in neurodegenerative diseases, making them more attractive therapeutic targets.

Beyond the Current Findings: What Comes Next?

The PanNDA project is an ongoing effort. Researchers plan to expand the atlas to include data from more brain regions, more diseases, and more individuals. They also aim to integrate PanNDA with other types of data, such as genomic and transcriptomic data, to create a more comprehensive understanding of neurodegenerative diseases. The team is working to develop new tools and algorithms to analyze the vast amount of data generated by PanNDA, making it easier for researchers to extract meaningful insights.
